    摘要:

    目的 对寡核苷酸芯片用于ABO血型基因分型的技术进行研究。方法 常规的酚/氯仿法提取标准血样基因组DNA,在ABO座位的外显子6和7区域各设计一对引物,经PCR扩增基因组相应区段并用Cy5-dCTP进行标记。设计寡核苷酸分型探针,将探针固定在APS-PDC法制作的DNA芯片上,用标记的PCR产物与之杂交,扫描仪对杂交结果进行扫描,Imagene软件对杂交图象进行分析。结果 本实验共检测了100份已知血型血样的ABO基因型,结果证明本实验研制的ABO基因分型芯片快速、准确、灵敏。结论 寡核苷酸DNA芯片用于ABO的基因分型与其他方法相比更具有灵敏、直观、高通量和集成化的优势。

    Abstract:

    Objection To develop an oligonucleotied chip for genotyping of ABO blood group.Methods The genomic DNA in standard blood samples was extracted by phenol-chloroform method.A pair of primers were designed according to the seguences of exon 6 and 7 of ABO locus.The corresponding seguences were amplyfied and the PCR products were labeled by Cy5-dCTP.The group-specific probes were designed and immobilized on DNA chips which were made by APS-PDC method.The labeled PCR products were hybridized with the probes ard the signals were scanned by scanner and analyzed by Imagene softwale.Results One hundred of samples which ABO blood group were known were determined by the oligonuleotide chip and both the accuracy and sensitivity were satisfied.Conclusion DNA chip is available for genotyping of ABO blood groups.Compared to other methods it is more sensitive and intuitionistic and has advantage of integration.
