Objective To deal with the questions in oligonucleotide chip,PNA probes were subsituted for DNA probes and PNA chips were prepared.By hybridizing with the ODN and HBV-DNA PCR fragment,the hybridizing conditions of PNA chip were discussed. Methods Both the DNA chip and the PNA chip were used to detect the single-base mutation.And the PNA chips also were used to detect HBV-DNA.Results Capability of recognization of PNA probed were stronger than those of DNA when they were used to detect the single-base mutation.The results about detection of HBV-DNA were identical when a kit was used.Conclusion The unique structure of PNA decides the advantages in hybridization such ad the high stability and strong speciality.
