    目的对聚苯乙烯板(PS)进行适当的修饰处理,以改善酶免疫分析的测定效果。方法以辣根过氧化物酶(HRP)为固相分子,用以观察酶与特异性底物之间的相互作用;用重组人增强肝脏再生因子(rhALR)、双链DNA(dsDNA)、心磷脂为包被抗原,以未氧化型与氧化型抗ALR为包被抗体,分别固定于不同修饰的PS板表面进行酶免疫分析,并与未经修饰处理的PS板作对比研究。结果生物分子的固定效果除与固相表面特征不同有关外,还与其本身的性质、所处的物理状态(包括分子的种类、结构、分子量大小和表面电荷等)密切相关。其中APTES与PLL修饰表面可显著改善rhALR、dsDNA、心磷脂抗原及氧化型抗体分子的固相化效果,提高酶免疫分析的吸光度值和测定敏感度。UV辐照PS板用于抗ALR和抗dsDNA抗体,醛基化表面(GA修饰)用于抗心磷脂抗体的测定亦可获得较高的特异信号显示与低背景噪音。AFM表征结果显示:疏水物理吸附的生物分子分布是随机和不规则的,且为聚集成团分布;而电荷引力与共价结合模式固定的生物分子空间取向与分布较为均一,可提高固定生物分子的数量和功能化保留程度。结论PS板经修饰处理后可改善对免疫分子的吸附能力,基于共价键和电荷引力结合模式可获得较为满意的酶免疫测试效果。

    Abstract:

    Objective To improve the measuring results of enzyme immunoassay (EIA) by appropriate modification of polystyrene microtitre plates (PS) with several different methods.Methods The interaction between enzyme and its substrate was investigated by using horseradish peroxidase (HRP) as immobilizing molecules.The coating antigen molecules,including recombinant human augmenter of liver regeneration (ALR),double strand DNA and cardiolipin,and the coating anti-ALR antibody molecules with different status (unoxidized and oxidized antibody) were immobilized on the modified PS plates respectively and investigated by EIA.A synchronous EIA was performed to compare the results by using untreated PS plates.Results Besides the feature of solid-supports,the immobilizing effect of biomolecules was strongly associated with their intrinsic characteristics and physical status,including the molecular classification and structures,molecular weight and electric charge on the molecular surface,etc.Among them,the surface modified by APTES and PLL improved significantly the solidifying effect of ALR,dsDNA,cardiolipin and oxidized antibody and enhanced the absorbance value and sensitivity of EIA.The EIA measurement using ultraviolet-irradiated plate for anti-ALR and anti-dsDNA antibody and aldehyde-surface modified by glutaraldehyde (GA) for anti-cardiolipin antibody presented higher specific signals and lower background noise.The results of atomic force microscopy (AFM) characterization demonstrated that adsorbed biomolecules based on hydrophobic interaction exhibited randomized and irregular distribution and clustered agglutination,whereas the dimensional tropism and performance of the molecules immobilized based on the manner of covalent bond and attractive linkage of electric charge evenly spread on solid supports and presented homogeneous distribution.The amount of molecules adsorbed on the surfaces and their function-preserving level were significantly increased.Conclusion The modified PS plates increase the capacity of immune molecules attachment.The satisfied performance and effectiveness of EIA on solid supports may be achieved by a given pattern in which covalent bond is combined with electric charge attraction.
