Abstract:Abstract: Objective:To study the effects of immune complexes (IC) in rheumatoid arthritis (RA) patients′ sera on the secretion of cytokines of fibroblast-like synoviocytes (FLS) detected by liquichip assay. Methods:The serum IC in the sera of RA patients who were positive anti-cyclic citrullinated peptide (ACCP) antibody and healthy controls were isolated by polyethyleneglycol (PEG) precipitation. The levels of ACCP-specific IC were determined by indirect ELISA. FLS collected from RA patients were cultivated in vitro and were stimulated with RA patients′ serum containing ACCP-positive IC or ACCP-negative IC, and healthy control IC respectively. After 24h of incubation, the cell supernatants were collected and the levels of IL-1β, IL-2, IL-6, IL-8, IL-10, IL-15, IL-17, TNF-α, GM-CSF, EGF, VEGF secreted by FLS were detected by liquichip assay. Results:The stimulations on FLS for 24h with RA serum containing ACCP-positive IC (6/10) and ACCP-negative IC (4/10) could significantly induce the secretions of GM-CSF, IL-6 and IL-8, compared with control group(P<0.05). The levels of GM-CSF and IL-8 in the supernatants of FLS stimulated with ACCPpositive-IC were significantly higher than those of ACCPnegative IC group(P<0.01). There were no significant differences of the other eight cytokines (IL-1β, IL-2, IL-10, IL-15, IL-17, TNF-α, EGF and VEGF) among the three experimental groups. Conclusion:ACCP-positive IC showed more efficiency on the induction of GM-CSF, IL-6 and IL-8 secretion from FLS.
