Abstract:Abstract: Objective:To detect Sluggene and its protein expression in the tissues of both cervical cancer and chronic cervicitis, and explore the correlation between Slug expression and the development of cervical cancer. Methods:The primary cancerous tissues from 31 patients with cervical cancer and the inflammatory tissue from 17 patients with chronic cervicitis were collected. The expression of Slug was detected by reverse transcriptionPCR (RT-PCR) and real-time RT-PCR. Slug protein levels were determined by western blot. The correlation analysis was conducted to assess the relationship between Slug expression and clinical pathological parameters, load of high risk human papillomaviruses (HPV). Results:The expression rate and relative expression level of Slug gene in cervical cancer tissues was significantly higher than that in chronic cervicitis tissues (χ2=9.872;U=140, P<0.01), which was verified by Slug protein levels determined by western blot (U=98, P<0.01). TheSlug expression in cervical cancer tissues was correlated to International Federation of Gynecology and Obstetrics (FIGO) stage. The positive rate of Slug gene in stage Ⅱ of cervical cancer was higher than that in stage Ⅰ (χ2=4.037, P<0.05). TheSlug expression was associated with the invasion of tumor cells, but no association with age of patients, tumor size, grade of tumor differentiation, lymph node metastasis and pathological type. A positive correlation between Slug expression and load of high risk HPV was observed in cervical cancer group (r=0.554, P<0.01). Conclusion:Slug expression in cervical cancer tissues was upregulated and associated with FIGO stage. Slug may play an important role in the pathogenesis of cervical cancer with synergistic effect of high risk HPV.
