    摘要:目的:探讨中国北方汉族人群TRIB1基因rs2235108多态性与2型糖尿病(T2DM)合并冠心病(CHD)的关系。 方法:用PCR限制性片段长度多态性(PCR-RFLP)检测了147例健康人对照组、96例T2DM组和75例T2DM合并CHD组TRIB1基因rs2235108多态性基因型和等位基因频率分布,分析基因多态性对T2DM和T2DM合并CHD的影响。 结果:我国北方汉族人群TRIB1基因rs2235108多态性CC基因型频率为81.8%,CT+TT为18.4%,C、T等位基因频率分别为90.5%、9.5%。3组研究对象的TRIB1基因rs2235108多态性基因型和等位基因频率分布差异无统计学意义(P>0.05)。3组内不同基因型间各项生化指标差异均无统计学意义(P>0.05)。logistic回归分析显示,年龄、HDL-C、高血压病史是T2DM合并CHD的独立危险因素。 结论:TRIB1基因rs2235108多态性与T2DM合并CHD无明显关联性,不是我国北方汉族人群T2DM合并CHD发病的独立危险因素。

    Abstract:

    Abstract: Objective:To explore the correlation between the polymorphism of TRIB1 gene rs2235108 and type 2 diabetes mellitus (T2DM), T2DM combined with coronary heart disease (CHD) in Han population of Northern China. Methods:Genotypes of TRIB1 gene rs2235108 polymorphisms and the distribution frequency of allele in 147 healthy individuals, 96 patients with T2DM and 75 patients with T2DM combined CHD were detected by polymerase chain reactionrestriction fragment length polymorphism (PCR-RFLP). The correlations between the polymorphism of T2DM gene, rs2235108 and T2DM, T2DM combined CHD were analyzed. Results:Genotypic frequencies of CC, CT and TT of TRIB1 gene rs2235108 were 81.8%, 17.3% and 0.9%, and the frequencies of C and T alleles were 90.4% and 9.6%, respectively. There were significant differences in the genotype and allele frequencies of TRIB1 gene rs2235108 between Han population of Northern China and the populations from other regions or races. However, there was no significant difference of genotype and allele frequencies of rs2235108 among the T2DM patients, T2DM patients combined with CHD and healthy controls (P>0.05). Likewise, there was no statistical difference of serum lipids, glucose and HbA1c levels among the 3 groups. Logistic regression analysis showed that age, HDL-C level and history of high blood pressure were the risk factors of T2DM combined with CHD. Conclusion:The polymorphisms of TRIB1 gene rs2235108 were not obviously correlated with T2DM combined with CHD, thus it should not be the independent risk factor of T2DM combined with CHD in Han population of Northern China.
