Abstract:Abstract:Objective:To determine the changes of IL-17 family cytokines in serum of rabbit model with skin graft after injecting recombinant human IL-10 (rhIL-10) and explore the effects of IL-17 family in the rhIL-10induced mechanism of reducing immune rejection. Methods:Rabbits with skin graft were divided into 6 groups, including low and high dosage of rhIL-10 groups, low and high dosage of cyclosporine A (CsA) groups, combined low dosage of rhIL-10 and CsA group and physiological saline group. Both rhIL-10 and CsA were intramusculary injected to the rabbits from the day before skin-graft for 10 consecutive days. Blood samples were collected on the day before skin-graft and the day 1, 4, 7, 14, 21 and 28 after skin-graft. The sera were separated and the levels of IL-17 family cytokines were determined by ELISA. Results:The levels of IL-17A, B, C, D, E and F after skin-graft were significantly higher than those before the operation, which reached the peak values on the day 14 and then gradually decreased. rhIL-10 inhibited the secretion of IL-17A, IL-27 and I-17F, but promote the secretion of IL-25. The inhibitory effect of rhIL-10 was positively correlated with the dosage of rhIL-10. Conclusion:IL-17 family cytokines may participate in the rejective reaction of graft and mediate the inflammatory reaction after transplantation.
