目的 对1例血型血清学正反定型不符的标本进行ABO血型鉴定。方法 运用血型血清学方法鉴定标本ABO血型;用PCR-序列特异性引物(sequence specific primer,SSP)基因定型和ABO基因直接测序的方法确定该标本的基因型。结果 血型血清学检测结果表现为B(A)亚型的特点;PCR-SSP分型结果定为B(A)04/O1型;测序发现标本ABO基因的第6和第7外显子存在变异:第6外显子存在261delG、297A>G;第7外显子存在526C>G、640A>G、657C>T、703G>A、796C>A、803G>C、930G>A,判断该样本血型为B(A)04/O01 血型。结论 对1例血型血清学初步鉴定为B(A)的标本,运用PCR-SSP法和ABO基因直接测序法确认其为B(A)04/O01型。
Abstract:
Objective To identify ABO blood group in a case of blood group serological inconsistences. Methods ABO blood group of the specimen was identified by blood group serological, and the genotype was determined by PCR-sequence specific primer (SSP) sand ABO gene direct sequencing. Results Serological test showed the characteristics of B(A) subtype. PCR-SSP test result was B(A)04/O1. Sequencing analysis indicated that the 6th exon had 261delG and 297A>G mutations and the 7th had 526C>G, 640A>G, 657C>T, 703G>A, 796C>A, 803G>C, 930G>A mutations, and the blood type of the sample was B(A)04/O01. Conclusion A case of blood group serological identified as B(A) was confirmed as B(A)04/O01 by PCR-SSP and ABO gene direct sequencing method.
