Abstract:Objective To investigate the correlations of Klebsiella pneumoniae carbapenemase (KPC)-2 and KPC-3 with main epidemic clones by analyzing the data of sequence types (STs) of global KPC-producing Klebsiella pneumoniae (K. pneumoniae). Methods The genomes of global K. pneumoniae were downloaded from NCBI and the distribution of KPC among these strains was analyzed. The genome STs of KPC-producing K. pneumoniae were determined by the multi-locus sequence analysis. Then, the distribution of KPC in different clone strains was analyzed, and the prevalence rates of main KPC variants such as KPC-2 and KPC-3 in the main epidemic clones ST11, ST258 and ST512 were investigated. Results A total of 1943 strains of KPC-producing K. pneumoniae were reported, and they contained 14 variants of KPC, mainly KPC-2 (959/1 943, 49.4%) and KPC-3 (952/1 943, 49.0%). A total of 115 STs were found, and ST258 (996/1 943, 51.3%), ST11(285/1 943, 14.7%) and ST512(259/1 943, 13.3%) were the major STs. Eighty-seven STs were distributed in 959 strains of KPC-2, and ST258 (413/959, 43.1%) and ST11 (274/959, 28.6%) were the dominant STs. Forty-five STs were distributed in 952 strains of KPC-3, and ST258 (570/952, 59.9%) and ST512 (259/952, 27.2%) were the dominant STs. The prevalence rate of KPC-2 in ST11 clone strains (274/285, 96.1%) was significantly higher than that in ST258 strains (414/996, 41.6%, χ2 = 43.819, P=0.000). The prevalence rate of KPC-3 in ST512 clone strains (259/259, 100%) was significantly higher than that in ST258 strains (570/996, 57.2%, χ2=206.645,P=0.000). Conclusion Globally, the prevalence of KPC is dominated by KPC-2 and KPC-3. The epidemic clone strains of KPC-2 are dominated by ST11 and ST258, and those of KPC-3 are dominated by ST258 and ST512. Strengthening the monitoring of these bacteria plays an important role in the prevention and control of nosocomial infection.
