摘要:目的 初步评估应用液相色谱串联质谱技术(LCMS / MS)对病原菌进行鉴定的可行性,为该技术临床应用提供实验依 据。方法 以全基因基础上的蛋白质组学库作为通用细菌鉴定数据库,用LCMS / MS技术对 41 株标准菌株以及临床分离的 22 株奈瑟菌和 46 株大肠埃希菌进行鉴定。结果 串联质谱技术鉴定病原菌在种的水平鉴定准确率为 100%。以 13 株标准 菌株样品稀释倍数和检测获得的肽段数量进行线性分析发现,二者之间具有很好的线性关系,相关系数均在 0.90 以上。结论 LCMS / MS 有望成为病原菌鉴定检测新方法。
Abstract:Objective To evaluate the feasibility of the identification for pathogens by tandem mass spectrometry (liquid chromatogra phytandem mass spectrometry,LCMS / MS)and to provides its experimental basis in clinical application. Methods A proteomic da tabase based on the whole gene was established as a general database for bacterial identification,and 41 standard strains,22 strains of Neisseria and 46 strains of Escherichia coli isolated from clinical samples were identified by the established LCMS / MS. Results The accuracy of the identification for pathogens at species level by tandem mass spectrometry was 100%. Based on the linear analysis of the dilution ratio of the samples from 13 standard strains and the detectable number of peptides,a good linear relationship was found be tween the two methods with above 0.90 of correlation coefficients. Conclusion LCMS / MS should be promising to become a new meth od for identification and detection of pathogens.
