    摘要:目的建立并优化一种基于微波消解结合离子色谱法同时测定血清中钠、钾,钙,镁4种阳离子的方法,并对其性能进行评价。方法﹐采用微波消解技术对血清样品进行前处理,使用离子色谱定量检测4种阳离子。参照美国临床和实验室标准协会(CL.SI)发布的EP6-A和EPI5-A3文件对建立的方法进行线性、精密度,加标回收率和正确度等性能验证。结果钠,钾、钙,镁4种离子的检测不精密度以变异系数(CV)表示,分别为0.10%~0.36%,0.15%~0.48%,0.22%~0.87%,0.21% ~0.73%。4种阳离子的加标回收率均在97.3%~103.0%之间。参加RELA2020和RELA2021计划的分析结果偏差分别为-0.57%~-0.14%,-0.84% ~ -0.16%、一1.82%6 ~-0.37%、-0.60% ~~0.34%,比对均通过。4种阳离子的检出限/定量限分别为0.0175/0.058 3 mmol/L,0.000 5/0.0017 mmol/L1,0.009 9/0.032 9 mmol/L,0.001 1/0.003 5 mmol/L,表明经前处理后,其他杂质对测定无明显干扰。结论﹐成功建立并优化了一种基于微波消解结合离子色谱法同时测定血清中钠、钾,钙,镁4种阳离子的方法。该方法精密度和正确度良好,操作简单,分析时间短,成本低,可用于临床血清电解质检测的质量改进。

    Abstract:

    Abstract: Objective To establish and optimize a method for simultanewus determination for the contents of sodium, potassium,calci-um and magnesium ions in serum using microwave digestion combined with ion chromatography , and evaltate its peformance.MethodsSerum samples were pretreated by microwave dligestion,and four cations were quantitatively detemined using ion chromatography.The linearity , precision, recovery and acuracy of the established method were verified with reference to the EP6-A and EPl5-A3 docu-ments isued by the Clinical and Laboratory Standarls Institute (CLSI).Results The imprecisions of deterninations for sotdium, po-tassium,calcium and magnesium ions expressed as oefficient of variation ( CV) were 0.10% to 0.36% ,0.15%e to 0.48%,0.22% to0.87% and 0.21% to 0.73% , respectively. The recoveries with adding standard of the four cations were between 97.3% anmd 103.0%.Referred to RELA2020 and RELA2021 , the deviations of the analysis results were -0.57% to -0.14% ,-0.84% to-0.16%,-1.82%6to -0.37% , and -0.60% to 0.34% for sodium, potassium, calcium , and magnesium , respectively , and all the comparisons were pae-sed. The limits of detection/ quantification were 0.0175/0.058 3 mmol/L,0.000 5/0.001 7 mmoL,0.009 9/0.032 9 mmol/L and0.001 1/0.003 5 mmol/L for the four cations,indicating that after pretreatment, no significantly interfere from other impurities for theresuls of determinmation was found.Conclusion A method for simultaneous determination of sodium , potassium, calcitm and magnesi-um ions in serum was successhully established and optimizedl using microwave digestion combined with ion chromatography. The methodshowed good precision anmd acuracy , simple operation , shot time consumption and low cost, thus it could be used for quality improve-ment of clinical electrolyte testing in serum.
