Abstract:Abstract: Objective To establish a quantitative method for the detection of human serum Dermatophagoides pteronyssinus-specific IgGbased on chemiluminescence immunoassay(CLIA),and evaluate its preliminary application. MethodsAn indirect CLIA based on magnetic nanoparticles was established for the quantitative detection of serum Dermatophagoides pteronyssinus-specific IgG. The re-action system was composed of streptavidin-labeled nano-magnetic particles,biotin-labeled D. pteronyssinus allergens, and alkaline phosphatase-laeldatimanatibdprcisininaangelimiofdtinndauacoftsalis method were evaluated. Results The coefficients of variation(CVs) of the precision of the established CLIA method were less than 10%.The linear range and LoD of the method were O.1-50 mg/L and O.1 mg/L, respectively. Twenty clinical serum samples were de-tected for Dermatophagoides pteronyssinus-specific IgG, by the Phadia AB IgG,assay and the established method, respectively,and the regression equation was y=1.684x-1.679 7 with a coefficient of determination(R2)of 0.947 1. ConclusionThe established CLIA methodcan accurately detect the level of human serumDermatophagoidespteronyssinus-specific IgGa,providing the experimental evidence for the precision therapy of related allergic diseases.
